Lantern Pharma (NASDAQ: LTRN), an AI-driven precision oncology company, reported updated Phase 2 HARMONIC trial data showing that LP-300, in combination with carboplatin and pemetrexed, demonstrated a progression-free survival (PFS) benefit that increased with treatment duration in patients with EGFR Exon 21 L858R-mutant non-small cell lung cancer (NSCLC) who had progressed following tyrosine kinase inhibitor (TKI) therapy.
The emerging data showed a median PFS of 8.9 months among L858R patients treated through up to six cycles, compared with 8.4 months across the overall L858R cohort. The company described the safety and tolerability profile as exceptionally clean, supporting a recently FDA-cleared protocol amendment extending LP-300 dosing from six to eight cycles.
This development is significant because it addresses a critical unmet need in NSCLC patients with the EGFR L858R mutation, who often develop resistance to TKI therapy. The HARMONIC trial is evaluating LP-300, a cisplatin/ethacraplatin analog, in never-smoker patients with relapsed advanced lung adenocarcinoma following TKI treatment. The ability to extend dosing without compromising safety could provide a meaningful therapeutic option for this patient population.
