Creative Biolabs has launched an integrated OMV and exosome-boosted CAR-T development platform, a vesicle-engineering approach intended to help researchers investigate strategies for overcoming the barriers that have limited CAR-T activity in solid-tumor models. The Shirley, New York-based biotechnology and contract research company announced the platform on October 27, 2026.
The announcement targets a persistent problem in immunotherapy research. CAR-T therapy has transformed outcomes in hematological malignancies, but its impact on solid tumors remains constrained by poor T-cell infiltration through dense tumor stroma, an immunosuppressive tumor microenvironment that drives metabolic exhaustion, and safety concerns such as cytokine release syndrome, along with antigen heterogeneity and escape. Conventional CAR-T approaches alone have struggled to surmount these barriers simultaneously.
The new platform integrates two complementary nanoscale vesicle technologies, bacterial outer membrane vesicles (OMVs) and mammalian-derived exosomes, into a unified, customizable pipeline for vesicle-enabled CAR-T research and tumor-microenvironment modulation. Each vesicle type addresses a distinct bottleneck, enabling researchers to evaluate complementary effects across multiple barriers.
“For years, the field has accepted that solid tumors require a fundamentally different approach to CAR-T therapy,” said a senior scientist at Creative Biolabs. “By integrating OMV and exosome engineering into a single platform, we’re giving researchers a dual-mechanism toolkit that doesn’t just chip away at these barriers—it enables researchers to investigate them in parallel, from stroma penetration to metabolic persistence.”
The platform’s OMV engineering arm functionally remodels the tumor microenvironment to promote CAR-T infiltration. Engineered OMVs can carry selected immunomodulatory or delivery payloads. A standout innovation is the BROAD-CAR system, in which OMVs transiently program tumor cells in vivo to display synthetic antigens recognized by a universal CAR-T construct, providing a preclinical strategy for evaluating antigen redirection across heterogeneous tumor models. OMV surface engineering with targeting ligands guides CAR-T homing precision, while detoxified LPS strains require characterization of immunostimulatory activity and endotoxin-related safety, a critical differentiator from synthetic liposome carriers that lack intrinsic immune-activating capacity.
The exosome engineering arm provides a biocompatible support system to sustain CAR-T activity within the suppressive tumor microenvironment. CAR-exosomes, which directly display functional CARs on their surface, deliver antigen-specific tumor killing without live cell administration. Selected exosome systems are being investigated for transport across biological barriers, including the blood-brain barrier, and can be engineered to carry cytokines, checkpoint inhibitors, and survival signals intended to modulate CAR-T exhaustion and persistence.
A third service line equips both vesicle types with customized therapeutic payloads. Small molecules, nucleic acids and proteins can be co-encapsulated within a single vesicle for multimodal synergy, while vesicle-mediated checkpoint blockade enables localized, sustained PD-1/PD-L1 inhibition directly at tumor sites.
The launch matters because it gives solid-tumor researchers a way to study stroma remodeling and persistence support in parallel rather than one barrier at a time. Creative Biolabs is offering the integrated OMV & exosome-boosted CAR-T development platform as a customizable pipeline that includes NTA sizing, high-resolution imaging and pharmacokinetic profiling.
